De groei/niet-groei interfase is de grens tussen condities die microbiële groei toelaten en condities die groei inhiberen. Modellen die deze interfase beschrijven, worden ontwikkeld in de discipline predictieve microbiologie om de veiligheid van levensmiddellen te voorspellen. De doeleinden van dit proefschrift zijn: ( i ) de grondige analyse en vergelijking van verschillende types van groei/niet-groei modellen voor zowel een monocultuur als een cultuur bestaande uit meerdere stammen, en ( ii ) de studie en het modelleren van de invloed van de celdensiteit op de groei/niet-groei interfase. Meer bepaald wordt de interfase van de pathogeen Listeria monocytogenes onder condities typisch voor mayonaise gebaseerde salades bestudeerd en gemodelleerd. Beide doelstellingen worden verwezenlijkt aan de hand van gedetailleerde datasets gegenereerd bij hoge celdensiteit en bij verschillende celdensiteiten. Om een groot aantal celdensiteiten tot op het niveau van de individuele cel te bekomen, wordt een verdunningsprotocol opgesteld. Een model dat dit protocol simuleert, wordt ontwikkeld om de celdensiteiten te schatten. De logistische regressiemodellen blijken het meest geschikt om de data bij hoge densiteit te beschrijven. Zij geven de geleidelijke verandering in groeikans geobserveerd in de interfasen, accuraat weer. Voor de monocultuur is het type afgeleid van een kinetiekmodel het meest geschikt aangezien zijn modelstructuur het minst beïnvloed wordt door anomaliën in de data. Er wordt onderzocht wat de beste methode is om de parameters van dit type te schatten. Voor de gemengde cultuur is het polynomiale type het beste. Alleen dit type is flexibel genoeg om de interfase van deze cultuur, die in een atypische richting buigt, te beschrijven. Om de invloed van de celdensiteit te modelleren, wordt de celdensiteit geïncorporeerd in de logistische regressiemodellen als extra verklarende variabele. Verschillende modellen die al dan niet gebaseerd zijn op de modellen voor hoge celdensiteit, worden opgesteld en uitgetest. De performantie van al deze modellen is quasi even goed. Zij voorspellen allen een sterke daling in groeikans bij dalende celdensiteit. Momenteel is er een voorkeur voor het model afgeleid van een kinetiekmodel omdat het het minst gevoelig is voor anomaliën. Tot besluit kan gezegd worden dat dit proefschrift belangrijke bijdragen levert tot het accuraat modelleren van de groei/niet-groei interfase en het eerste werk is dat grondig de celdensiteit in deze modellen incorporeert. The growth/no growth interface is the boundary between conditions that allow microbial growth and conditions that inhibit growth. Models that describe this interface are developed in the discipline of predictive microbiology in order to predict the safety of food products. The objectives of this dissertation are to ( i ) thoroughly analyse and compare the performance of different types of growth/no growth models for a monoculture as well as a culture consisting of several strains, and ( ii ) study and model the influence of the cell density on the growth/no growth interface. In particular, the growth/no growth interface of the pathogen Listeria monocytogenes under conditions typical for mayonnaise-based salads is investigated and modelled. Both goals are accomplished on the basis of detailed data sets generated at high cell density and at several cell densities. To obtain a large number of cell densities up until the level of the individual cell, a dilution protocol is developed. A model simulating this protocol is constructed to estimate the cell densities. The logistic regression models are found most suitable to describe the high density data. They accurately describe the gradual change in growth probability observed in the growth/no growth interfaces. For the monoculture, the type derived from a kinetic model is most adequate since its model structure is least influenced by anomalies in the data. It is investigated which method is most appropriate for the estimation of the parameters of this model type. For the mixed strain culture the polynomial type is the model type of preference. Only this type is flexible enough to describe the interface of this culture which bends in an atypical direction. To model the influence of cell density, the cell density is incorporated in the logistic regression models as an additional explanatory variable. Several models of which some are based on the models built for high cell density, are constructed and tested. The performance of all these models is closely similar. They all reflect the observed strong decrease in growth probability with decreasing cell density. At the moment, the model derived from a kinetic model is preferred since this model is simple and least sensitive to anomalies. In conclusion, this dissertation contributes to the accurate modelling of the growth/no growth interface and is the first work to profoundly incorporate cell density into these models. Voedselveiligheid is wereldwijd van primordiaal belang. Er treden immers regelmatig voedselgebonden ziekten op. Het aantal voedselvergiftingen kan verminderd worden door een goede kennis van de groei van schadelijke micro-organismen in voedingsmiddelen. Er kan dan immers voorspeld worden onder welke condities een bepaald voedingsmiddel best bewaard wordt, enz. In het domein van de predictieve microbiologie worden er modellen ontwikkeld die de groei van micro-organismen onder verschillende omgevingscondities voorspellen. Een subgroep van predictieve modellen zijn de groei/niet-groei modellen. Die modellen voorspellen de groei/niet-groei grens, i.e., de grens tussen omgevingscondities die groei toelaten en omgevingscondities die groei inhiberen. In dit project worden groei/niet-groei modellen ontwikkeld voor het micro-organisme Listeria monocytogenes in gekoelde mayonaise gebaseerde salades zoals, bijvoorbeeld, surimi-salades. De voor- en nadelen van een aantal modeltypes worden onderzocht en vergeleken. Een belangrijk innoverend aspect van dit project is dat de groei/niet-groei grens niet enkel bij hoge contaminatiegraad bestudeerd en gemodelleerd wordt, zoals vaak in de literatuur gebeurt, maar ook bij lage contaminatiegraad, wat meer realistisch is.
